If you forget to put in conditioner, the water won't hurt your turtle. You can just leave your turt in the water and add the conditioner later. Conditioner is good - chlorine in the water (which conditioner neutralizes) can cause mild eye irritation to a turtle, and good conditioners contain other things such as ammonia neutralizers that are beneficial. However, lack of conditioner won't hurt a turtle.
If there are also fish in the tank, the chlorine in unconditioned water could kill them, however. Fish aren't like turtles - they breath the water and are a lot more sensitive to chemicals than turtles.
When you said you put a lot of the conditioner inside the tank, do you mean you put the conditioner in after you filled the tank with water? When you say you put in a lot of conditioner, did you put more than the directions on the box call for? Be sure and only put in as much conditioner as the box specifies - too much conditioner can irritate turtle eyes.

spotsmama's right. it would not really hurt the turt, but could hurt the fishes (if you have some) in the tank. i have to put some conditioner in the tank before i put everything back inside.
i just finished cleaning my tank 10 minutes ago (my shirt's still wet and everything...). and i almost forgot to put the conditioner in the water... good thing i saw this post. heh heh heh... thanks for reminding...
